Abstract
Transplantation of germ line of rams could open new paths in the field of animal reproduction. Until now, researchers have developed techniques very expensive and difficult to apply practically.
The aim of this study is to develop a method that is simple, fast and economically sustainable that allows to perform an inoculum of mesenchymal stem cells in testes of Sarda rams.
In first experiment was tested on rams Sarda the possibility of eliminating the resident germ cell line by the use of a vaccine. The vaccine consists of an analogue of GnRF, that in recent years is used in pig breeding industry to prevent the castration of pigs.
The second trial, however, was to develop a surgical technique that would permit access to the mediastinum testis via extra-testicular testis-rete.
In exp.1, 10 Sarda lambs were divided into 2 groups;group1(n=5) was injected with anti-GnRH Vaccine and group2 (n=5) was the control.
In exp.2, Mitotracker green was injected into the extra-testicular rete testis of 5 Sarda rams and the diffusion of the dye into the testis was evaluated.
The result's showed that the anti-GnRH vaccine inhibited the production of testosterone and reversibly depleted the germ cell line.
Also the surgical access to seminiferous tubules into the extra-testicular rete didn’t cause testicular damage and the animals resumed normal sexual activity.
