Mammary tissue biopsies were taken on d -5, -2, 1 and 6 relative to a single 500 mg dose of bST (Posilac®, Monsanto, St. Louis, MO). Total RNA was isolated, amplified, converted to cDNA, labeled with Cy3 or Cy5 dye, and hybridized to the NBFGC bovine microarray (Michigan State University). Comparisons were made using a modified loop design with each cow on each day represented once. Milk yield was increased (30%; P < 0.05) by d 4 and remained elevated through the end of the study. Milk fat, protein and lactose content were not affected (P > 0.1).
